Education Essay

The most prominent example of a formal setting in the book is school. However, Scout does not learn much from school. This can be examplified by the fact that when Scout is able to read better than the teacher, Miss Caroline Fisher had expected, Miss Caroline Fisher told her to tell her father to stop teaching her how to read. Instead of helping Scout to improve her reading, Mrs Fisher is impeding Scout’s learning process. This shows that schools do not cater to children of different abilities and Scout’s learning is restricted in school. Hence, school is not the only place where a child learns. Jem and Scout learn moral values from Atticus. This is examplified by the many values Atticus teaches them such as moral courage and to stand up for what one believes is right. Atticus brought his children up to treat the blacks and the whites as equals. When Scout tells Atticus to send Calpurnia away, Atticus told Scout that they could not survive a day without Calpurnia and he told Scout to â€Å"mind† her. Scout learnt that she had to treat Calpurnia (a Negro) like how she would treat a White and to be respectful towards Calpurnia. Jem looks up to Atticus and tries to emulate him. This is because Atticus is a good role model and a good father. Jem learns from Atticus to have the moral courage to fight for what is right. This is evident as Atticus went all out to fight for Tom Robinson as he believed that Tom was innocent. Atticus did this although he knew that he would face severe criticism from the people in Mycomb and that he would put his life and his children’s at risk. From the example he sets, Jem is able to learn to have the moral courage to fight for what is right. Hence, Jem and Scout do learn in informal settings. Jem also learnt to have the courage to do what one decides to do. This is evident from Jem’s encounter with Mrs. Dubose. Mrs Dubose is a morphine addict and she tried very hard to get rid of her addiction. Mrs. Dubose sets an alarm clock and made Jem read to her as he was angry at what she said and cut off the top of her bushes. When the alarm clock rings, it would mark the end of Jem’s reading time. During this period of time when Jem is reading, Mrs Dubose will refrain from taking morphine. She used this method to try and get rid of her morphine addiction. To further elaborate, Mrs. Dubose was very courageous to try and get rid of her morphine addiction as it was a very painful process. Mrs. Dubose was almost dying but she wanted to leave the world beholden to nothing so she tried her very best to get rid of her addiction and displayed a strong sense of courage. Jem learnt from Mrs. Dubose that no matter how painful the process is, one should have the courage and determination to accomplish it. Hence, Jem do learn in an informal setting. Jem and Scout also gained knowledge from a new experience that Calpurnia showed them. This is evident as Calpurnia showed them how it was like in a Negro church. By visiting their church, they learnt about the difference in the conditions of the Negroes’ church and their church. They also learnt more about Tom Robinson’s case when Scout asked Calpurnia why the pastor demands a donation from everyone to help Tom Robinson’s wife provide for her kids. From these, the children were somewhat more aware of the discrimination happening in Maycomb.

In this context, J.  P. Morgan is the bright example of the way mutual funds are used to reduce the negative impact of financial crisis and to overcome the difficulties faced in tough bond markets. J. P. Morgan has probably been the first to use mutual funds as the instrument of protection against the negative impacts of financial crisis. In his article, Michael Pollock (2009) sheds the light on the way J. P. Morgan Strategic Income Opportunities fund helps the company deal with tough bond markets. It appears, that the fund â€Å"has few restrictions typical of bond funds that are marketed to general public† (Pollock, 2009); as a result, it is better equipped to help investors survive through the difficult financial times. The fund functions according to a predetermined set of principles, of which putting money only into places where potential profits overweigh potential risks is probably the most important. The mutual fund at J. P. Morgan does not avoid keeping a portion of assets in cash, so that investors can materialize their investment opportunities when the right moment comes. Short selling is just another instrument the fund uses to generate additional profits; Pollock (2009) also notes that short selling is becoming a widely spread investment tool among bond funds. The list of investment instruments J. P. Morgan uses to manage its mutual fund is not limited to short selling and cash operations. Here, investors are also given a chance to make short borrowings and then to sell these borrowed shares; â€Å"investors can also make similarly bearish bets by buying credit-derivative instruments whose value increases if the price of an underlying corporate bond declines† (Pollock, 2009). To a large extent, the fund relies on the whole set of quantitative techniques that work to identify significant investment opportunities. The fund is actively involved into managing long-term high-yield corporate securities and nonagency mortgage-backed bonds. As a result, the fund has been able to achieve the total return rate of 4. 3% this year (Pollock, 2009). Does that mean that beyond using mutual funds as investment targets and the sources of additional profits, companies can also utilize the benefits of portfolio investment to protect themselves from external crisis threats? There is no definite answer to that question, but J.  P. Morgan obviously tries to change traditional opinions about investment options available to consumers. The truth is that everything we currently know about mutual funds does not make them look as an ideal investment solution. Given that mutual funds are not usually guaranteed by the FDIC and are not insured by any government agency; that mutual funds’ past performance is not always indicative of its future positive prospects; and that to be a member of a mutual fund also means to bear certain costs associated with investments, the whole picture of a mutual fund does not look as much attractive. However, where J. P. Morgan was able to reach the point of total return rate of 4. 3%, investors may have some sort of confidence that the company will pursue the same set of investment principles, being extremely cautious in its investment options and using the mutual fund as an effective means of anti-crisis protection. Conclusion Mutual funds are included into the list of the most widely used investment options. It appears, however, that mutual funds can also be successfully used to protect companies and investors from the negative impact of the financial crisis. Despite the costs investors have to carry as well as unpredictability of external environments, which mutual funds cannot control, the latter remain relatively stable and non-volatile means of dealing with tough bond markets.

The term ‘decree’ means a rule of law that is issued by someone in authority within the legal system. Generally, decree is issued by the head of state. Although, a decree does not constitute all the features of an order, this particular degree represents certain specific procedures. The main intention of this d ecree was to ensure equal employment opportunities in NOPD and thereby eliminate all sorts of racial discrimination practices. Obviously, the decree brought considerable modifications in the NOPD’s promotion procedures mainly with the aim to increase the opportunities for the advancement of African-American officers. The proposed decree enforced the creation of supernumerary positions. However, a group of officers who are not African-American raised voice against this provision. In order to comply with the framed stipulation structure, PANO informed NOPD its need to maintain additional regular sergeant positions. The City has made certain adjustments in the administration of sergeant appointment in order to give equal priority to African-American officers and other officers. As a result of this practice, the city could maintain a total of 16 new sergeants, out of which six African-American officers were from Band 6 and ten non African-American officers were from Band 5. The P ANO and other 24 police officers who were not African-American alleged that the City dealt with certain transfers and promotions in a manner that violated the intention of the decree. In fact, all the 24 police officers who opposed the move were already in Band 5 of the Commission’s promotional register, and hence, according to them, promoting the Band 6 African-Americans adversely affected the employment opportunity of those 24 individual officers. The plaintiffs argued that the transfers and promotions that were exercised on 31st December 1993 were a blatant violation of the equal protection and due process clauses of the Fourteenth Amendment to the United States Constitution, and Article I, Section 3 of the Louisiana Constitution of 1974† (Findlaw). The district court found that the promotions and transfer practices of the City on 31st December 1993 were against the decree and fundamental laws of the US constitution. Although, the district court gave the City an oppo rtunity to find an effective remedy that would correct the identified violations, the City could not frame a potential remedy within the specified date. Most importantly, the situational theory require leader to adjust his behavior and leadership style to features of followers and situation. Therefore, it might be important to look at the four styles described by Hersey and Blanchard to identify the situations that any leader might have to change his style or behavior. There are situations where leader might have to be flexible with his followers by giving them less responsibly and delegation of tasks due to their lack of knowledge/experience, or if they are newly hired etc. the leader in this case might need to utilize the telling style whereby there is more of directing and controlling of leader to his followers. In this behavioral style, leaders role involves telling his follower on what and how to perform their tasks step by step. In my working experience, for example, I tend to use this style when training people whom are newly hired or inexperienced and require a lot of assistance or hints when in doubt. I might sometimes need to spill out the responsibilities or duties for perform to make sure that they are still on focus. Knowing that differences between tasks and relationships behaviors as described in Hughes et al (1996), I agree that the telling style does have high task and low relationship with followers because followers in this stage are unable to perform any delegated tasks and that means should be giving detailed instructions. Another situational and behavioral style is the selling style whereby the leader is acting both as a director and supporter toward his followers. This style might be more effective if followers are delegated toward some responsibility and have an adequate knowledge of performing their tasks. The leader in this case is only giving directions and guideline when any deviation is observed. In term of skills and ability, the leader is also to promote his followers skills throughout. Because of that, it ca be said that in this behavioral style, the leader is coaching i.e. there is higher tasks delegation and higher relationship with followers. In the participating style, the leader exercises low tasks delegations but high relationship with followers. This is because in this style the main role of leader is to facilitate the delegated tasks to his followers by showing his knowledge or skills on how best can this be done. Bearing in mind the level of communication is high in this style which enables follower to share in the decision making process as applicable. This behavioral style might best be used is when followers are starting or completing a job (Hughes et al, 2006). The last situational style is the delegating style whereby there is little two side communication between the leader and his followers (low-task- low relationship). This style might be implemented when leader is aware of his followers knowledge, ability, confidence, experience and most importantly taking higher responsibilities. To this extend, followers are able to direct them selves with no control from their leader. Although, leaders in this style are like an observers whereby they have less influence on their followers and less control. However, leader might also need to consider followers development level/their readiness level and competence. Followers competence might include their knowledge, skills and ability to assess the work outcome. Therefore, leader might have to respond or change his behavior in such way that fits the situation. He can classify his people to stages such as:Â · People are able and willing to take tasks (competent). Â · People are unable and unwilling to take tasks (incompetents)Â · People are unable but willing to take tasks (unskilled). Â · People are able but unwilling tasks (unmotivated). So to summarize, it can be seen that the situational leadership theory presented by Hersey and Blanchard focuses on the followers behaviors and define what style is best to suit any situation. Also, it highlights the level of responsibilities delegated as well as the degree of relationship between leaders and followers. Put into practice, situation leadership theory is basically effective with any situation. Moreover, it seems that the situational theory answer the question that leader might need to change their behavior and be flexible with followers as that depends on situation and circumstances. In other words, leaders might need to adjust them selves all the time depending on the working environment and follower level of readiness (Norris, 1992). And as the Hersey and Blanchard theory, they should always change their personality or be able to identify clues in their environment and adapt their leader behavior to meet the needs of their followers in any particular situation (). Vecchio and Norris agrees that ,even with good diagnostic skills, leader might need to be flexible in changing their behavior and adapt their leadership styles that meet that demand of both the followers and environment. There are several factors that can influence a business, these factors include Internal and External influences. Internal influences include factors the business itself may have some control over, whilst on the other hand factors that the business can not control such as laws and regulations are External influences. Importance of business Business is like the blood that flows through our bodies, with out business our lives would be almost impossible. Business plays many different roles in our everyday lives. Most people either own or work for a particular business. Businesses make it possible for the community and even further places such as overseas countries to receive and sell goods and services. A business can be anything that provides goods and services to the community such as corner shops providing customers with goods and sweets and they can also be larger companies such as Nike selling shoes and clothing all over the world. Businesses allow for our economy to grow and become stronger, with out a strong economy a country can not run as smoothly as preferred. One of the main reasons why businesses are so important in our everyday life is due to the fact that businesses provide millions and millions of people with job opportunities. If businesses did not exist people will not be able to work and earn money for e veryday living. There are certain factors that can influence a business. These factors include internal and external influences. Internal influence Internal influences are influences that a business has some control over, such influences include product, location, management, resource management and business culture. 2 Product influences: These influences affect a large majority of the internal structures and operations of a business. Depending on the types of goods and services the business produces will depend on the internal structure. If the goods being produced require certain equipment, these needs must be catered to so that the business may go on with its production. Each type of business has a different set of product influence, for example a service provider will require less preparation, whilst on the other hand a manufacturer will need more services to produce the goods sold. The size of a business can be determined by the different types of goods and services produced and also the availability of technology. So from this we understand that the amount of goods and services produced by a business depends on the size of the business. Information resources are all the actions in which the business takes to research and learn more about their situation for eg sales reports. Physical resources include all the equipment and tools necessary for running the business. Financial resources are basically what the business depends on to buy and help support their business, for example to buy new machines. 5 Management influences: There is no doubt that over time businesses and the way in which businesses are run have dramatically changed. Management has been a part of business that has been changed in an effective way. In the past there were many levels of management and hierarchy in a business that new ideas and issues had to go through, but now due to many factors such as the pressure of competitiveness from globalisation the way in which businesses are run have been altered. These days there are much fewer levels that new ideas and issues have to go through, this will result in quicker and more efficient decisions that in the long run will allow the business to focus on the way it runs. This allows businesses to adapt quicker and more efficiently to the ever changing needs and wants of consumers because there are fewer managers in which the process of decision making must go through. 6 Business culture: In reality every business has its own type of business culture, these are all the values, beliefs and ideas that are agreed upon by all employees and managers of the business. The culture of a business is shown in the way in which the business acts and performs. Business culture is usually shown in the rules and regulations of a business such as the way staff are expected to dress and behave. Culture has been proven to be a necessary tool that must be used by businesses to achieve the best out of its staff members and customers. So basically it is essential for businesses to be affected by internal influences in a positive way. External influences External influences are factors that a business may have little or no control over, such factors include: Economic, financial, geographical, social, legal, political, institutional, technological, competitive situation and markets influences. 8 Economical influences In Australia and the rest of the world, the economy experiences economic cycles. These cycles include booms and busts (recession). Basically during an economic boom the economy experiences higher levels of employment, they may be so confident in their sales that they may increase the price of items to gain higher profits. Also during booms wages of employees tend to increase as more profit is being made by the business. However the economic cycle does have its down side, such as when the economy is experiencing a recession period. An unfortunate reality of recessions is that employment rates decrease due to the fact that the business is not making enough sale profits to keep their employees. Another effect of recession is that business may not be able to raise the price of their products because people are not purchasing so they may have to even lower prices to continue to compete in the market. Even better, now due to globalisation, Australia is now able to trade goods overseas, further more improving the financial sustainability of the country. 10 Geographical influences Geographical influences impact businesses in many ways. Australia is located in the Asia-pacific region, the economical statuss in these surrounding countries are very important in the interest of Australia. One other major social influence is the influence of of our environment which is basically wearing away day by day, businesses must always consider what is in the best interest of the environment, for eg not giving out plastic bags after a sale. One final issue is the issue of family-work practices, as women give birth they must leave work, and this may result in the business losing an employee, affecting in the production of the business. Generally the people of a society expect that businesses may run under the rules and guidelines provided by the law. Businesses must be aware of all the laws so that they may not breach them and pay the penalties. 13 Political influences Even though political influences may not be the most affective factors onto a business, they still have a pretty heavy impact. For eg whenever a new election Is held, the new government may introduce new policies. A policy that was introduced in 2000 was the goods and services tax, this was a 10 percent tax on the supply of most goods and services used in Australia, this affected businesses in a way that they had to collect taxes basically on behalf of the government. However deregulation has also been introduced in the picture in the past few decades, this is the removal of government regulation in industry, and this takes pressure off businesses and allows for a greater range of competitiveness. 14 Institutional influences Three institutional influences on business are government, regulatory bodies and trade unions and employer associations. Government: Three levels which are federal, state and local. Federal government deals mainly with issues such as tax, making sure that the taxes are being paid and also things such as how the businesses are running in according to the customs regulations. State governments deal with lower case issues such as workers rights and responsibilities such as OHS requirements and making sure businesses are abiding by the state laws and trade practices Local government deals with issues such the condition of building and making sure of improvements, parking rules and fire regulations Regulatory bodies; These bodies observe and understand the way that businesses deal with certain issues and how the business treats its community and consumers. Examples of regulatory bodies include: the Office of fair trading and the Australian competition and consumer commission. (ACCC) Other institutional influences are Trade unions and Australian stock exchanges, these groups have aims such as improving working conditions and pay rates. 15 Technological influences Technology without a doubt has had probably one of the most affects on businesses. With new technologies being invented and discovered every day this can only help improve productivity and efficiency for businesses. Technology has allowed for robotic machinery to be used in productivity which in the long run cheapens the cost of productivity as no employee or has to be paid. Robots also help to reduce the amount of boring and repetitive jobs that some humans may have to do. Also communication technology has been improving rapidly over the past few years making it even easier to communicate with suppliers and consumers, with out them actually being there. Businesses must strive to use the available technology to the best of their ability so that they may keep up or even be ahead of competitors. 16 Competitive situation influences These influences may be very helpful and useful to both consumers and producers, for eg when two businesses are competing to be the market leader, they will lower costs of their stock to attract customers. As a result they will be making more sales and more importantly a profit. These competitive situations also benefit consumers because with competition in a market there will be a greater range of goods available for consumption, this allows the consumer to get a wider variety of choice when considering the good or service they want. External influences can be both positive and /or negative for example, competitiveness in the market allows for a variety of options for consumers to choose from but contrasting the effects of external factors such as geographical issues involving the weather can have a negative affect, for eg flooding in farms or drought seasons. Gene Therapy Methods and Delivery Systems Genes the units of heredity â€Å"There can be little doubt that the idea of ‘the gene’ has been the central organizing theme of twentieth century biology† philosopher and biochemist Lenny Moss claimed in 2003. A gene is the basic unit of heredity in a living organism. Genes hold the information to build and maintain their cells and pass genetic traits to offspring. In general terms, a gene is a segment of nucleic acid that, taken as a whole, specifies a trait. The biological entity responsible for defining traits was termed a gene, but the biological basis for inheritance remained unknown until DNA (Deoxyribonucleic acid) was identified as the genetic material in the 1940s. In cells, a gene is a portion of DNA that contains both coding sequences that determine what the gene does, and non-coding sequences that determine when the gene is active (expressed). When a gene is active, the coding and non-coding sequences are copied in a process called transcription, producing an RNA (Ribonucleic ac id) copy of the genes information. RNA can then direct synthesis of proteins via the genetic code. In other cases, the RNA is used directly, for example, as part of the ribosome. The molecules resulting from gene expression, whether RNA or protein, are known as gene products, and are responsible for the development and functioning of all living things. Every cell requires a host of genes that act as blueprints of all the proteins essential for its proper functioning. DNA is a linear polymer of deoxynucleotide monomers. Chemically speaking, it has a double helical structure of two polynucleotide chains held together by hydrogen bonds between the complementary base pairs of the nucleotide strands. Each nucleotide in DNA is comprised of three components, a heterocyclic base, a sugar (2-deoxyribose) and a phosphate group. The nucleotides in a polynucleotide chain are connected through phosphodiester bonds. The nitrogenous bases are of two types, namely, purine based adenine (A) guanine (G), and pyrimidine based cytosine (C) thymine (T). In DNA, base pairs form only between A T and G C and thus the base sequence of each single strand can be deduced from that of its complementary strand. Gene Therapy: Molecular Bandage? Gene therapy is believed by many to be the therapy of the twenty first century because it aims to eradicate cause rather than symptoms of diseases by delivering a normal functioning copy of the mutated gene and its associated regulatory elements into the cell nucleus (1-3). It is a technique whereby an absent or a faulty gene is replaced by a working gene, so that the body can make the correct enzyme or protein and consequently eliminate the root cause of the disease. A potential approach for treating genetic disorders is gene therapy. The most likely candidates for future gene therapy trials will be single gene disorders like, cystic fibrosis, hemophilia, familial hypercholesterolemia, ADA deficiency, Gaucher disease, alpha-1-antitrypsin deficiency etc. Apart from these monogenic disorders, gene therapy also holds the potential of treating acquired diseases such as cancer, by inhibiting oncogene expression or by restoring tumor suppressor genes or through immunomodulation (i.e. by i ncreasing immune response to tumor antigens). Cardiovascular diseases too remain as one of the most â€Å"promising† targets for gene therapy because of the ready accessibility of the vascular system for gene transfer (4). Types of Gene Therapy: Depending on the type of cells into which genes are transferred a process popularly known as â€Å"transfection†. Gene therapy can be broadly classified into two types: Somatic cell Germline gene therapy. Somatic Cell gene therapy: This type of therapy involves the transfection of somatic (non-reproductive) cells especially of those tissues in which expression of the concerned gene is critical for health. Expression of the introduced gene relieves/eliminates symptoms of the disorder, but this effect is not heritable. Germline gene therapy: This type of therapy involves gene transfer into reproductive cells (egg or sperm cells). Here germ cells are modified by the introduction of functional genes, which are ordinarily integrated into their genomes. It would change the genetic pool of the entire human species, and future generations would have to live with that  change. Depending upon the method of correcting the faulty gene, gene therapy can be classified into the following categories: Gene augmentation therapy: This type of therapy is the most appropriate one for the treatment of inherited diseases caused by the loss of a functional gene. It involves supplementing the body cells with a functional copy of the lost gene so that the missing protein is expressed at sufficient levels in the body. It is only suitable if the pathogenic effects of the disease are reversible. Gene inhibition therapy: Its aim is to introduce a gene whose product inhibits the expression of the pathogenic gene or interferes with the activity of its product (5). Suicide gene therapy: This method is best suited for a disease like cancer where the aim is to eliminate a certain population of cells. It involves the transfection of such cells with a suicide gene, whose product is toxic. The suicide genes should be appropriately targeted to avoid widespread cell death (6). Depending upon the mode of delivering genes into a patient’s body, gene therapy can be classified into the following two types: Ex vivo gene therapy: In ex vivo gene therapy, gene transfer occurs outside the patient’s body. This is again sub-divided into two types viz, Autologous Non-autologous. Autologous gene therapy involves the transfection of cells derived from the patient followed by the re-introduction of these cells into the patient’s body. Non-autologous gene therapy involves the transfection of cells not derived from the patient’s body. In vivo gene therapy: In this method the therapeutic gene is directly introduced into the body by injection or by inhalation with the help of a suitably designed vector. Gene Delivery Vectors: Key to Success in Gene therapy Gene therapy, as a novel therapeutic modality, holds enormous promise for the treatment of a multitude of human diseases. However, till date it has failed regrettably in spite of more than 1500 clinical trials completed or currently underway around the world. The primary reason for the failure of the clinical success of gene therapy is the lack of efficient gene delivery agents, commonly referred to as transfection vectors. However, since the biological cell surfaces are negatively charged (due to the presence of glycoproteins and glycolipids containing negatively charged sialic acid residues on cell surface), spontaneous entry of polyanionic naked genes (DNA) into body cells is an inefficient process. Hence â€Å"transfer vehicle† or a â€Å"vector† in needed to condense the macromolecular DNA and to help it in crossing the plasma membrane barrier. Again delivery of therapeutic DNA to the desired body tissue is important to overcome adverse affects. In other words, the problems of developing clinically viable gene therapy methods and designing safe efficient gene delivery reagents are inseparable: shortcomings in one is going to adversely affe ct the success of the other. Hence, realization of the full potential of gene therapy will depend, in a major way, on the future development of safe and efficient gene delivery vectors. The Ideal Vector!!! A â€Å"perfect† or an â€Å"ideal† vector would resemble a traditional pharmaceutical and should have the following characteristics: (a) should be capable of efficiently delivering to its target an expression cassette carrying one or more genes of the size suitable for clinical application, (b) must not elicit an immune response, (c) should not induce inflammation and thus be safe for the recipient, (d) can be produced in bulk at an acceptable cost with reproducibility, (e) should be stable on storage, and finally, it should express the gene (or genes) it carries for as long as required in a strictly regulated manner. No single vector currently available has all these desired properties and each vector presently in use has its own pros and cons. However, it is important to realize that there cannot be a â€Å"universal† vector, optimally useful for all gene therapy applications. This is due to the fact that each disease will have a unique set of technical requirements, and the â€Å"perfect† vector for a specific disease should be optimized in accordance with these requirements. For example, some diseases will require local delivery of the transgene (e.g., ischemia, retinitis pigmentosa, parkinson’s disease, etc.) while others likecancer and atherosclerosis necessitate systemic delivery. In some cases, only a transient, short-lived gene expression will be needed (e.g., therapeutic angiogenesis, cancer) while in monogenic disorders, such as familial hypercholesterolemia, hemophilia and SCID a long term (sometimes life long) gene expression is mandatory (1). The future clinical success of gene therapy will certainly depend on the uphill task of designing â€Å"tailor-made† vector systems for the treatment of specific diseases. The efforts to design a â€Å"perfect vehicle† for the membrane-impermeable DNA have so far led to the development of many methods based on the principles of biology (viral vectors), physics (microinjection, electroporation, particle bombardment, hydrostatic pressure, and ultrasound) and chemistry (synthetic vectors like cationic lipids polymers). Each of these methods has its intrinsic advantages and disadvantages. Viral Vectors: Nature’s Own Infecting Vehicles Viruses have evolved specific mechanisms through the course of evolution to deliver their genetic material into host cells and then hijack the cell’s biosynthetic machinery to produce new viral particles (7). Thus, owing to their natural ability to infect cells, they can be used as vectors in gene therapy by replacing the genes that are essential for replication phase of their life cycle with the therapeutic genes of interest. Majority of the clinical trials currently underway around the world are based on the use of mainly five categories of viruses, namely, retrovirus, adenovirus, adeno-associated virus, lentivirus and herpes simplex virus. Retroviruses: These are a class of enveloped viruses containing a single stranded RNA molecule (approximately 10 kb). Inside the host cell, the RNA is reverse transcribed into double stranded DNA, which in turn integrates into the host genome and is expressed as viral proteins (8). These are the most promising and widely used viral vectors used for gene therapy applications to date. Advantages: Rapidly dividing cancer cells can be targeted by using these viruses. Enters into cells efficiently and gives long lasting gene expression due to stable integration. Disadvantages: Only infects dividing cells, capable of producing tumorigenic mutagenesis due to random integration, unable to deliver larger genomic sequences. Again, it can insert the genetic material of the virus in any arbitrary position in the genome of the host- it randomly shoves the genetic material into a chromosome. Adenoviruses: These are the second most commonly used viruses for gene delivery. They carry a double stranded linear DNA chromosome of approximately 36 kb. Unlike retroviruses, adenoviruses deliver their genetic payload outside the chromosome and are thus less likely to disrupt the cell’s genome (9). But it is immunogenic and may cause inflammation and tissue damage. Adeno-associated viruses (AAV): They contain a single stranded DNA of approximately 4.7 kb surrounded by a protein coat (10) and can integrate at a specific site in human chromosome 19. AAV does not contain any viral genes and contains only the therapeutic gene and it does not integrate into the genome. It requires co-infection with a â€Å"helper† adenovirus for propagation.
